Tower of God centers around a boy called Twenty-Fifth Bam, who has spent most of his life trapped beneath a vast and mysterious Tower, with only his close friend, Rachel, to keep him company. When Rachel enters the Tower, Bam manages to open the door into it as well, and faces challenges at each floor of this tower as he tries to find his closest companion.

In September 1944, allied forces led by the 501st Joint Fighter Wing "Strike Witches" successfully eliminate the Neuroi threat from the skies of the Republic of Gallia, thus ensuring the security of western Europe. Taking advantage of this victory, allied forces begin a full-fledged push toward central and eastern Europe. From a base in Petersburg in the Empire of Orussia, the 502nd Joint Fighter Wing "Brave Witches," upon whom mankind has placed its hopes, flies with courage in the cold skies of eastern Europe.

The heroine, Komori Yui, was a normal teenage girl. In her second year of high school, Yui's father, a priest, moves somewhere else for his work. As a result, Yui is forced to move to a new town and arrives alone at a mansion she was told will be her new home. As she approaches the front door it begins to rain. Because no one answers her knock, she reaches for the knob, but the door swings open of its own accord. Yui enters the mansion to find no one in sight. As she starts to explore her immediate surroundings, she finds a young man(Ayato) asleep on a couch. He wakes up as she approaches him and promptly pins her down. As she begins to panic, five other young men gradually appear. Yui notices something different about all of them. She discovers that all six of them are brothers but by three different mothers, later revealed to be Beatrix (mother to Shu and Reiji), Cordelia (mother of the triplets Ayato, Kanato, and Raito), and Christa (mother to Subaru). However, these young men don't seem quite human. In fact, they all turn out to be vampires.

In an alternative 19th century, Allen Walker is an Exorcist, a soldier of God who hunts devils and demons called "Akuma". Allen is soon sent, by his Master Cross Marian, to England, where the Headquarters of the Exorcists, The Dark Religious Organization, resides. Together with the other Exorcists, who each posses the power of "innocence", Allen must track down Akuma all over the world and kill their master, The Millennium Earl, before he destroys the world! However, things are much more then they seem. There are higher-level Akuma who posses immense power and intelligence. Among these are those who are part of Noah's Family, a group descended from Noah himself. These individuals have allied themselves with the Earl, and combined with the decreasing number of Exorcists who are part of the Dark Religious Organization, Allen and his fellow Exorcists are in for a tough ride with a bleak future!
